First I sketch a character out and then ink it on the computer then color it then shade it. I will often reuse alot of my art such as characters in the same position. So that makes it easier. I make it all in layers so all the pieces of the characters are moveable and then I can draw the piece I need and it fits in place. Just about all my work is done on the computer because I like the ability to draw freehand well.
Most people use programs like gimpshop or photoshop and/or similar programs.

It definitely looks vector based, and I would guess he does this in Flash.
